高产精氨酸脱亚胺酶菌株的复合诱变选育及其发酵工艺优化

摘要:以粪肠球菌SK 32.001为出发菌株,经过化学诱变和常温室压等离子体诱变,平板初筛和摇瓶复筛,获得了高产精氨酸脱亚胺酶菌株A3。经过发酵调控工艺条件优化进一步提高了该菌株的产酶能力。结果表明,在3 L发酵罐水平下,以蔗糖为碳源,牛肉膏为氮源,溶氧值控制在20%,诱变菌株A3的酶活为3.12 U/mL,比原始菌株提高了162.18%。

Breeding of High Arginine Deiminase Producing Strain by Compound Mutagenesis and Its Fermentation Process Optimization
Abstract:To improve the arginine deiminase producing ability, Enterococcus faecalis SK 32.001 was mutated using chemical method coupled with atmospheric room temperature plasmas. A high arginine deiminase producing strain A3 was obtained by plate initial screening and re-screening in shake flask fermentation. Then the enzyme producing ability of the strains was further improved by optimizing fermentation conditions. The results showed that the enzyme activity of the mutant strain A3 was 3.12 U/mL with sucrose and beef extract as carbon and nitrogen sources, respectively, and with the dissolved oxygen value was controlled at 20%. Compared with the original strain, the enzyme activity was improved by 162.18%.
