In this study, different molecular weight CM-chitosans were prepared and the effects on the growth and collagen secretion of normal skin fibroblasts and keloid fibroblasts were investigated in vitro. CM-chitosan promoted the proliferation of the normal skin fibroblast significantly but inhibited the proliferation of keloid fibroblast. The higher CM-chitosan concentration had a higher initial effect and the lower CM-chitosan concentration had a longer affecting time to the normal skin fibroblast. The lower molecular weight CM-chitosan had significant twofold activities. The CM-chitosan could reduce the ratio of type I/III collagen in keloid fibroblast by inhibiting the secretion of collagen type I; and had no effect on the secretion of types I and III collagen in the normal skin fibroblast. r 2002 Elsevier Science Ltd. Chitosan was modified by coupling with linoleic acid through the 1-ethyl-3-(3-dimethylaminopropyl)- carbodiimide-mediated reaction to increase its amphipathicity for improved emulsification. The micelle formation of linoleic acid-modified chitosan in the 0.1 M acetic acid solution was enhanced by O/W emulsification with methylene chloride, an oil phase. The fluorescence spectra indicate that without emulsification the self-aggregation of LA-chitosan occurred at the concentration of 1.0 g/L or above, and with emulsification, self-aggregation was greatly enhanced followed by a stable micelle formation at 2.0 g/L. The addition of 1 M sodium chloride promoted the self-aggregation of LA-chitosan molecules both with and without emulsification. The micelles of LA-chitosan formed nanosize particles ranging from 200 to 600 nm. The LA-chitosan nanoparticles encapsulated the lipid soluble model compound, retinal acetate, with 50% efficiency.

Carboxymethyl chitosan (CM-chitosan) was prepared by chemical reaction with monochloroacetic acid under various conditions, and the chemical structure was analyzed by IR and NMR. The water solubility of the CM-chitosans had close relationships to the modifying conditions and the degree of carboxymethylation. The CM-chitosans, prepared at temperatures of 0-10 8C were soluble in water. But the CM-chitosan prepared between 20 and 60 8C were insoluble in the water at near-neutral pH. The water insolubility of CM-chitosans at various pHs varied with the degree of carboxymethylation. The increase in reaction temperature increased the fraction of carboxymethylation and increased the insolubility at lower pHs; the increase of the ratio of water/isopropanol in the reaction solvent decreased the fraction of carboxymethylation and increased the insolubility at higher pHs.

通过玻璃流延法制备不同分子质量的壳聚糖及羧甲基壳聚糖膜,以体外培养的人皮肤成纤维细胞作为对象,利用膜的浸渍液培养及膜表面直接培养法研究比较了两种多糖膜的细胞相容性。实验发现两种多糖膜的浸渍液对细胞均无毒性效应,生物安全性是小分子质量膜好于大分子质量膜,羧甲基壳聚糖膜好于壳聚糖膜。皮肤成纤维细胞在壳聚糖膜上的生长受到抑制,生长一段时间后细胞有聚集和脱落现象,而羧甲基壳聚糖膜上细胞能很好地贴附、生长,没有聚集和脱落现象。结果表明羧甲基壳聚糖膜具有比壳聚糖膜更优越的细胞相容性。

以羧甲基壳多糖(CMCH) 为材料,研制出一种用于动物细胞培养的新型微载体CX-2。对该微载体的色泽、干燥失重、比表面积、悬浮密度、直径、吸水量、溶胀度、粒度分布、表面结构、机械强度、稳定性等理化指标进行了测定,结果表明,CX-2 是一种性能良好的微载体。细胞学实验表明,BB 细胞能较好地黏着于CX22 微载体上,在24~72h 期间细胞生长增殖速度快,120h 时细胞已呈现多层次生长状态。再生后的CX-2 ,可继续用于BB细胞培养,效果无明显变化。结果初步表明,CX-2 为新型的、适于动物细胞大规模培养的、可再生利用的微载体。

用分步流延干燥法制备了一种由壳聚糖(CS) 、聚乙烯醇( PVA) 、羧甲基壳聚糖(CM-CS) 构成的新型人工皮肤医用材料——壳聚糖复合不对称膜(C-P-C 膜) ,其上层为CS 层,中间为PVA 层,下层为CM-CS 层。研究结果表明,可通过调节CS、PVA、CM-CS 的加入量来控制复合不对称膜各层的厚度和膜的水蒸气通透性, C-P-C 膜具有良好的透光性和吸水率。生物相容性实验发现,C-P-C 膜是无毒材料,不会引起创伤感染,随着植入C-P-C膜时间的延长,组织炎症反应明显减轻,6 周时,炎症浸润程度低于手术缝合线。C-P-C膜能够明显地促进创面愈合,能有效地密封出血创面且具有显著的止血作用。

利用生物化学和细胞培养方法对研制出的新型羧甲基壳多糖微载体CX-2 的生物亲和性、细胞贴壁及生长和扩展特性进行了研究。研究结果显示:CX-2 在pH4~7.2 范围内对小牛血清和人血清有较强的选择性吸附作用,表现出良好的生物亲和性;BB (Bulltrout Branchia) 细胞贴壁率、贴壁速率、生长速度和扩增培养效果表明:CX-2 微载体具有良好的细胞相容性,细胞能快速贴壁、生长和扩增;Vero 、BHK细胞在CX-2 微载体上贴壁、生长和扩增与BB 细胞基本一致,表明该微载体具有广泛的适应性。实验结果显示:CX-2 不仅具有优良的理化性能,同时具有良好的生物亲和性和细胞相容性,是一种应用前景广阔、实用性强的新型微载体。

Chitosan microcarriers (100-200μm) were prepared by the methods of emulsification and ethanol coagulant. It has smooth surface and was stable in phosphate buffer solution (PBS) of at pH 7.2 in the treatment of temperature 120 ◦C and pressure 150 kPa. The chitosan microcarriers showed molecular affinity to the bovine serum proteins at pH 7.2. The adsorptive capacity of the microcarriers to the serum albumin was 6.8 mg protein/g chitosan bead. The chitosan microcarriers were found to have good biocompatibility and no cytotoxicity to both human and mouse fibroblasts in tissue cell culture. The fibroblasts well adhered on the smooth surface of the chitosan microcarriers and grew in high cell density. The results suggest a good potential of the chitosan microcarriers as a wound-healing biomaterial.
