林洪,男,1962年生,工学博士、教授、博士生导师,山东省学术带头人。从事水产品加工与食品安全性的教学与科研工作。现任中国海洋大学生命科学与技术学部副主任、山东省水生生物制品安全性重点实验室主任。全国水产专业标准化技术委员会水产品分技术委员会副主任委员。近5年发表学术论文35 篇,其中SCI收录6篇,主编学术专著一部。申请国家发明专利5项。承担国家、省部级科研项目近十项。主持和参与水产品标准制修订和审阅评定工作,累计完成30余项。指导博硕士生20余人。1999获山东省科技进步二等奖(7位),获2002山东省科技进步三等奖(1位)。2002年获优秀硕士论文指导奖。

对紫贻贝、栉孔扇贝、褶牡蛎、杂色蛤、缢蛏、毛蚶六种贝类的磷脂作了比较研究,对它们的出肉率、总脂、中性脂、极性脂、磷脂及其组分、脂肪酸的组成做了检测分析。结果表明,六种贝类的磷脂特性有一定的差异,牡蛎中磷脂酰肌醇含量特别高。心磷脂只在牡蛎和毛蚶中被检出,毛蚶中只含EPA,而无DHA。六种贝类共同特点是磷脂酰胆碱与磷脂酰乙醇胺之和在70%左右。

从生物化学和物理学两个方面比较了养殖对虾和海捕对虾肌动球蛋白的特性,研究了加热对蛋白的影响。结果表明:加热后两种对虾蛋白质的溶解度和Ca2+2A TPase活性随温度的升高而下降,Mg2+-ATPase活性的变化与Ca2+2A TPase的稍有不同。总巯基数从40℃开始略有下降,未结合巯基数先升后降。疏水性随温度升高而增大。ATP感度在常温时几乎没有变化,而35℃后有明显下降。所有指标的比较均说明,对虾肌动球蛋白对热反应强烈的温度始于30~40℃,且养殖对虾的热稳定性更差。

综述国内外对褐藻岩藻聚糖的化学组分、结构和生物活性的研究进展。其化学组成L-岩藻糖和硫酸基团为主,由于来源不同而含有不同比例的糖醛其他单糖组分。采用化学与物理方法分析结构,L-岩藻糖可以以α(1→3)、α(1→2)和α-(1→4)的方式连接成主链,并且石油同位点上有硫酸基团的单取代或双取代。这种组分和结构得褐藻岩藻聚糖具有抗凝血、抗肿瘤、抗氧化、抗病毒等多种生活活性。

木瓜蛋白酶经戊二醛固定于自制的甲壳质上制成固定化木瓜蛋白酶,其水解甲壳胺的最佳条件是:45℃,pH 4.0的1%的甲壳胺溶液(醋酸缓冲液配置),酶/底物比1∶3.5(质量比)。震荡反应24h后,用超滤器分离不同分子量的甲壳胺低聚糖产物,结果产物中分子量小于10000 的甲壳胺低聚糖得率约为45.55%,分子量小于5000的约为34.0%,优于游离木瓜蛋白酶水解甲壳胺的效果。此固定化酶可反复使用至少10次以上,总反应时间达192h,提高了酶的使用效率和酶解效果。

从8种载体材料中选取自制的氨基含量约2.878mmol/g的甲壳质作为固定化载体,采用吸附交联法,以戊二醛为交联剂,对木瓜蛋白酶进行固定化。17.5mg/g的固定化酶载体比例,pH6.5,5℃下,先吸附10min,再以0.7%的戊二醛终浓度交联12h,所得固定化酶酶活回收可达52.0%,酶活力为3.54U/g。固定化酶在65℃以下,溶液酶在55℃以下稳定;固定化酶在pH7.0以下稳定,溶液酶在pH6.0以下稳定;5℃条件下,固定化酶贮藏半衰期为183d;以酪蛋白为底物,固定化酶的操作半衰期可达27d。用固定化酶水解甲壳胺,产物分子量小于10000的甲壳胺低聚糖得率约45.55%。

The phospholipids of mussels (Mytilus edulisl Linne) from the coast of Qingdao were extracted, fractionated and analysed over a 12 month period. Thecontents of total lipids, neutral lipids, polar lipids and phospholipids were meausred. The composition of phosholipids was determined by high-performance liquid chromatiorgraphy, and their fatty acid composition was analysed by gas chromatorgraphy. Thephospholipide contert ranged from 3.6 to 6.4 g kg-1 soft tissue. Pe (phosphatidyl ethanlaminc) and PC (phosphatidyl chline) were the major constituents. C16:0, C20:5 and C22:6 wree the major fatty acids. C20:5 (5.25-23.10%) and C22:6 (6.05-20.42%) varied regularyly with the seasonal factors, Their total amounts were high from January to Juhne, which wouldbe an optimal time for the utilisation of trhe phospholipids of mussels.

Muscle residue depletion of chloramphenicol (CAP), sulphamethoxazole (SMZ) and oxytetracycline (OTC) following oral administration was evaluated in shrimp, Penaeus chinensis under field conditions. Three groups of shrimps were cultured in tanks filled with seawater and fed a commercial medicated diet containing 2000 mg kg 1 CAP, SMZ and OTC, respectively, twice daily for 3 days. Sampling was conducted at different intervals (0, 0.5, 1, 1.5, 2, 3, 4, 8, 10, 12, 16, 24, 36, 48, 72, 96, 120, 144, 168 h) after the cessation of medication. Drug analysis was carried out by HPLC. The elimination half-life (t1/2) of CAP was 10.04 h, the elimination half-life (t1/2) of SMZ was 5.68 h and that of OTC was 16.12 h. If the EU MRPL value of 0.3 ng/g for CAP and MRL value of 0.1 Ag/g for SMZ and OTC quoted for muscle from finfish is extended to shrimp muscle, extrapolation of the data indicates that it would be passed after a 139.7 h (95% CI=132.0-144.4 h), 30.6 h (95% CI=27.2-33.1h), 90.3 h (95% CI=87.9-92.5h) withdrawal period for CAP, SMZ and OTC in shrimp muscle, respectively.

Immobilized papain was used to prepare chitosan oligomers and the optimum conditions were as follows: NaAc-HAc buffer system, pH 4.0, temperature 45℃. After 24 h reaction with stirring, nanofiltration and ultrafiltration were used to desalt and isolate chitosan oligomers into different molecular weight fractions respectively. The yield of chitosan oligomers with molecular weight under 10,000 was 49.55% to total substrate, while that of between 600 and 2000 was 11.07%. The fractions 01, 02 was analyzed by Sephadex G-50M gel filtration chromatography and flight mass spectrograph was employed to analyze the fraction O3. The results showed immobilized papain could depolymerize chitosan successfully, and separating the product by ultrafiltration membrane was feasible. Further study revealed that the immobilized papain could be repeatedly used at least 10 times. An amount of 1 mg free papain and mmobilized papain could produce 2.57 and 17.34 mg chitosan oligomers, respectively. The results indicated that the depolymerization efficiency of immobilized papain was obviously higher than that of free papain.

A low molecular weight sulfated polysaccharide (L-A) was prepared by mild acid hydrolysis of crude fucoidan (F-A) from Laminaria japonica. In comparison with F-A, L-A had a lower proportion of fucose residues, but a similar proportion of sulfate. The galactose content of both fractions was relatively high, especially for L-A (57%). The antioxidant properties of the two polysaccharides were studied using two low-density lipoprotein oxidation systems. L-A had a stronger effect against low-density lipoprotein oxidation in both systems. F-A inhibited the AAPH-induced low-density lipoprotein oxidation, but had little effect on the CuCC-induced system due to its large molecular mass. The active sulfated fraction L-A, containing alactose, mannose and fucose (about 9: 2: 2) is reported here for the first time.
