摘要

Anovel chemiluminescence (CL) flowsensor for in vivo on-line monitoring of glucose in blood by microdialysis sampling is described in this paper. The sol-gel method is introduced to co-immobilize horseradish peroxidase (HRP) and glucose oxidase (GOD) on the inside surface of the CL flow cell. The CL detection involved enzymatic oxidation of glucose to d-gluconic acid and H2O2, then H2O2 oxidizing luminol to produce CL in presence of HRP. The perfusion rate of the dialysate was 5.0ml/min. The dialysate sample volume was 20ml, and the sample throughput was 15h−1. The variation of glucose level in blood of the rabbit was monitored after the administration of glucose to demonstrate the favorable resolution and reliability of the system for in vivo on-line monitoring.

摘要

A novel chemiluminescence (CL) flow biosensor for hydrogen peroxide is described. It is prepared by immobilizing HRP with the sol-gel method on the flow CL cell, and by electrostatically immobilizing luminol on a strongly basic anion-exchanger resin. Hydrogen peroxide is sensed by the CL reaction with HRP sensitive membrane and luminol bleeding from the ion-exchanger column with immobilized luminol by hydrolysis. 摘要

A flow-injection chemiluminescence CL.biosensor for the determination of total D-amino acid is described in this paper. D-amino acid oxidase was immobilized onto amine-modified silica gel via glutaraldehyde activation and packed in a column. The other analytical reagents, including luminol and ferricyanide, were electrostatically co-immobilized on an anion-exchange column. D-amino acid was sensed by the CL reaction between hydrogen peroxide produced from the enzyme column and luminol and ferricyanide, which were eluted from the ion-exchange column through sodium phosphate injection. The linear range of D-alanine concentration is 1.1×10-6 to 1.1×10-4mol/l and the detection limit was 4.5×10-7mol/l (3σ). The proposed method has been successfully applied to the determination of D-amino acid in human serum.
