The total synthesis of hirsutellide A 1 was described. The linear hexadepsipeptide precursor 2 was synthesized in 45% yield from N-Boc-Me-Gly by three coupling reactions with DCC, HATU and BOP–Cl, respectively. Macrocyclization was successfully performed on the fully deprotected amino acid 14 with BOP–Cl in 15% yield and with FDDP in 22% yield.

【期刊论文】Selective synthesis of cis-2,6-dimethylpiperazine catalyzed by a Cu-Cr-Fe/γ-Al2O3 catalyst

A continuous process for the synthesis of cis-2,6-dimethylpiperazine was described in this paper. The cyclization of N-β-hydroxypropyl-1,2-propanediamine proceeded effectively over a Cu-Cr-Fe/γ-Al2O3 catalyst in a fixed-bed reactor. Cu0 and Cu+ were believed to be the active sites of the catalyst. Fe was found to play an important role on the catalyst activity and stability. Effects of solvents, temperature and hydrogen pressure were examined; solvents were found to be very important for this reaction. Remarkable differences were found between the catalysts used in water and those used in dioxane by the XPS and SEM characterizations. The loss of Fe and some carbon deposit on the catalyst surface were detected and supposed to be the main reasons of the catalyst deactivation in water, whereas the catalyst showed an excellent service life in dioxane. Finally, cis-2,6-dimethylpiperazine was obtained with a total yield of over 70% under the optimum reaction conditions. Thus, a highly selective process for the synthesis of cis-2,6-dimethylpiperazine was established here.

Racemization of R-2-amino-1-butanol proceeded effectively over a fixed-bed Raney cobalt catalyst in a continuous fixed-bed reactor under the hydrogen pressure in the range of 2.0–6.0MPa. The performance of a fixed-bed Raney cobalt catalyst was compared with that of a pulverous Raney cobalt catalyst. The catalysts were characterized by X-ray photoelectron spectroscopy (XPS), X-ray diffractometry (XRD), environment scanning electron microscopy (ESEM) and N2 adsorption measurement (BET). The activity and stability of the fixed-bed Raney cobalt catalyst and the effects of concentration, temperature and hydrogen pressure were examined. Racemic 2-amino-1-butanol was obtained with a total yield of over 76% under the optimum reaction conditions. Thus, an effective process for the racemization of R-2-amino-1-butanol has been established.
