摘要

利用派罗宁B-黄色组分诱导孢粉素产生荧光的特异性,观察了水稻M-101品种以及它的无花粉型胞核雄性不育(GMS)突变体I-15系花药绒毡层解体过程中孢粉素的行为。M-101品种花药绒毡层的解体过程大体可分为两个阶段;在小孢子发育期,绒毡层主要分泌孢粉素;在花粉发育期,绒毡雇主要降解非孢粉素物质。I-15系花药绒毡层的解体过程虽然出现了一系列异常现象,但绒毡层解体的起始时阃和阶段顺序性与M-101品种相似。对绒毡层解体过程孢粉素行为的有关事态及其与花粉发育的关系等问题进行了讨论。

【期刊论文】SSR标记鉴定栽培稻杂种F1花粉愈伤组织基因型的偏态分离

摘要

分别对栽培稻FI花粉不育基因S-a、S-6和S-c单座位内互作杂种Fl进行离体花药培养,利用SSR分子标记鉴定了花粉愈伤组织的基因型。结果表明,不同座位F1产生的携带花粉育性基因Si或Si的花粉形成愈伤组织的能力偏离1:1的分离比,与自然条件下F2群体偏离方向不一致。携带不同花粉不育基因座位的F1产生花粉愈伤组织基因型的偏离方向有差异,表现为S-a和S-c单座位内互作杂种F1的花粉愈伤组织偏向Si,Si:Si比例分别为1:4.81和1:1.96;S-b单座位内互作杂种F1花粉愈伤组织偏向Si,Si:Si比例为1:0.35。研究培养条件对偏态分离的影响,发现预冷处理可明显提高偏态分离的程度,而花药培养基类型及诱导培养时间的长短不会改变偏态分离的方向。

摘要

The cytoplasmic male sterility (CMS) of wildabortive (WA) cytoplasm has been widely used for breeding hybrid rice. Two restorer genes for the CMS have been found by traditional genetic analysis. To tag the restorer genes we used a set of near-isogenic lines (NILs) of Zhenshan 97 carrying different genotypes for fertility restoration from IR24, to perform RAPD analysis. From the survey of 720 random primers, six RAPD markers were identified to be associated with Rf-3. Three of these OPK05-800, OPU10-1100 and OPW01350, were mapped on chromosome 1. Two Fa populations from the crosses between Zhenshan 97 A and a near-isogenic restorer line ZSR21 and between Zhenshan 97 A and IR24 were used for mapping Rf-3. The three RAPD markers and three RFLP markers, RG532, RG140 and RG458, were found to be closely linked to Rf-3 in the two F2 populations. The same location of RJ-3 was also found in a BC1 population from the cross of IR58025 A//IR36/IR58025 B. At the RG532 locus, different alleles were found between two CMS lines, Zhenshan 97 A and IR58025 A, and between two restorer lines, IR24 and IR36. The use of these molecular markers closely linked to Rf-3 in facilitating the development of hybrid rice is discussed.

Lu Y.-G. and RUTGER J. N. Anther and pollen characteristics of induced genetic male sterile mutants zn rice (Oryza sativa). ENVIRONMENTAL AND EXPERIMENTAL BOTANY 24, 209--218, 1984. Spikelet fertility, anther length, anther width, anther dehiscence, pollen stainability and number of pollen grains per anther were studied in 11 genetic male stcriles and their parent cultivar M-101. Four lines set some seed under bagged conditions, indicating that some self pollination occurred in these lines. Anther length was near-normal in several lines, while anther width was universally reduced. Seven lines had indehiscent anthers. Pollen stainability and number of pollen grains per anther ranged from zero to near-normal. One line showed anther and pollen characteristics similar to the fertile parent, except that the pollen of the sterile line failed to germinate. Based on the combined anther and pollen characteristics, the I 1 lines were classified into four types: stainable pollen abortion type (SPA), partial pollen abortion type (PPA), complete pollen abortion type (CPA), and no pollen type (NP). The CPA type, which is relatively easy to identify visually, and which shows appreciable outcrossing, appears to be the most useful type for male sterile facilitated population improvement schemes.
